Microbiology Analysis and APG
APG recently sent two chemists to water and wastewater microbiology training presented by EMD Chemicals in State College, Pennsylvania.
During the training, different aspects of the analysis for microbiology were discussed. One topic of interest was the different types of culture media available. The different types of media can be divided into the following types: Selective, Differential, Enrichment, Agars, and Broths. The type of testing will determine what media to use.
Another topic discussed was the different types of test being performed. The main tests are Most Probable Number (MPN) and Membrane Filtration (MF). MPN analysis of drinking water is by a series of ten test tubes. Each tube containing Lauryl Tryptose Broth (LTB) is inoculated with ten milliliters of sample. The tubes are incubated at 35 C for 24 hours. A positive result is indicated by the presence of gas inside the Durham tube. MF analysis of drinking water is done by filtering 100 milliliters of sample through a 0.45 micron filter. The filter is then placed into a Petri plate containing a medium necessary for growth. The plates are inverted and incubated for 24 hours at 35 C. A positive result is indicated by the presence of red colonies with green metallic sheen.
