DMR-QA Microbiological Samples

Complete Article from Edition 43 APG eNewsletter

Article written by Sharie McManaway , Production Supervisor, APG

Microbiological samples have been added to the DMRQA analyte list for study 26.  APG has two studies that meet the DMRQA deadlines.  Our DMRQA specific study will ship on July 10, 2006 to be received July 11, 2006 and analyzed on July 12, 2006.  Our July WP Microbiological study also may be used for DMRQA compliance, shipping on July 31, 2006, received on August 1, 2006 and tested on August 2, 2006.

APG has received many questions concerning the preparation and testing of these samples as they are new for many DMRQA participants.  Here is a brief description about what you will receive:  Our samples are provided as lyophilized samples that require an addition of phosphate buffer solution and incubation to reconstitute.  The buffer solution and pipette are provided with the sample.

Included below, there are some recurring answers to several, frequently asked questions.

- Samples must be incubated at 35 degrees C.

- The one milliliter added to the vial for reconstitution does not have to be exact, the pipette is not
...volumetric.

- If the buffer solution is frozen upon arrival, thaw at room temperature.

- Samples must be prepared on the Wednesday after arrival.  Samples analyzed on any other day
...may be an outlier in the study.

If you receive a Quality Control sample along with your unknown, the samples must be run on the
days indicated.  Certificates of Analysis will be sent to your laboratory a few days after the analysis
date; usually 2-3 days.

The sample product range is 20- 2400 CFU/100mL, or 20-2400 MPN/100mL.  Dilutions should be
prepared to capture values throughout the product range.  i.e. If plated at the following dilution, the results should be within the range specified as follows.
